The colorimetric quantification of proteins presents several limitations according with the biological sample. To quantify proteins directly into cultures of microorganisms is necessary to validate the method of your choice,
to guarantee that reagents and other compounds accumulated during growth do not interfere with the reaction. This technical note reports the interferences observed during protein quantification of nitrogen fixing bacteria gown into semi-solid media when glucose or gluconic acid was used as carbon source. The results shown here illustrate the limitations of methods considered validated and recommended when certain modifications are introduced
or occur during the execution of an experiment.
